That is a tough one. On paper - both phones are quite similar. The MyTouch 4G offers HSDPA+, which is great if you live in an area that has this upgrade. I've played with the MyTouch 4G, and would have a hard time picking between the two. The MT4G is heavier, and has a faster processor, but lacks the rugged features of the Defy.
As for the UI - I'm actually not a fan of either, I use Launcher Pro to replace Blur and Sense as I prefer my Android devices to be as clean as possible.
