This is a long-time Las Vegas practice and successful fairly often. It is well worth a try, because the room (or view) you'll get is more than a $20 per night upgrade and most people stay in Las Vegas more tha one night.
The key is to put your ID and credit card on the check-in couner with the bill folded under them (but the denomination visible) and immediatly ask them if their is a possibility to be placed in one of their nicer room. (They will understand what you are doing/saying).
For Bellagio, Venetian and Wynn Las Vegas you may want to use a $50 bill.
You really have nothing to lose. If they decline, they aren't rejecting you. They simply aren't in a position to help you out (hotel is full, perhaps someone in upper management is cracking down a bit on the practice).
Vegas is about taking a couple of risks. This is probably one of the better ones in town because you have nothing to lose!
